Effect of different nitrogen sources on the cellular form of Trigonopsis variabilis.

چکیده
Triangular cells are formed during early logarithmic growth of Trigonopsis variabilis. Good triangularity is obtained with methionine, proline, hydroxyproline, or alanine as a nitrogen source.

Induction of the d-Amino Acid Oxidase from Trigonopsis variabilis.
Induction of the d-amino acid oxidase (EC. 1.4.3.3) from the yeast Trigonopsis variabilis was investigated by using a minimal medium containing glucose as the carbon and energy source, (NH(inf4))(inf2)SO(inf4) as the nitrogen source, and various d- and dl-amino acid derivatives as inducers. The best new inducers found were N-carbamoyl-d-alanine, N-acetyl-d-tryptophan, and N-chloroacetyl-d-(alph...
متن کاملInhibition of Recombinant D-Amino Acid Oxidase from Trigonopsis variabilis by Salts
Inhibition of recombinant D-amino acid oxidase from Trigonopsis variabilis (TvDAAO) activity in the presence of different sodium salts and potassium chloride is reported. A competitive inhibition pattern by sodium chloride was observed, and an inhibition constant value of K(i) = 85 mM was calculated. Direct connection of NaCl inhibition with FAD cofactor dissociation was confirmed by measuring ...
متن کاملOptimization of media composition for D-amino acid oxidase production by Trigonopsis variabilis using biostatistical analysis.
D-amino acid oxidase (DAAO) is biotechnologically relevant enzyme that is used in various food and pharmaceutical industries. DAAO from the yeast Trigonopsis variabilis is an important agent for use in commercial applications because of its high activity with cephalosporin C and is reasonable resistant to the oxidants O2 and H2O2 byproducts of reaction. In this study, response surface methodolo...
متن کاملKinetic mechanism of D-amino acid oxidases from Rhodotorula gracilis and Trigonopsis variabilis.
The reaction of two D-amino acid oxidases from the yeasts Rhodotorula gracilis and Trigonopsis variabilis with the substrates alanine and valine in their 2-1H and 2-2H forms was studied employing the stopped-flow spectrophotometric technique. The turnover numbers at infinite substrate and oxygen concentrations were: 20,700/4,250 and 1,730/360 ([2-1H]/[2-2H]alanine and valine, respectively) for ...
متن کاملEnhancement of Trigonopsis variabilis D-Amino Acid Oxidase Overproduction in Fed-batch Cultivation of Escherichia coli
D-Amino acid oxidase is a key enzyme in the production of semisynthetic cephalosporins. The Trigonopsis variabilis D-amino acid oxidase with 12amino-acid peptide at N-terminus was expressed in Escherichia coli BL21(DE3), in a series of short fed-batch cultivations. The strategy of gradual induction and gradual feeding with D-methionine during expression resulted in active enzyme production twic...
